Phosphorylated Antibodies . All rights reserved.G. Ren and G. LiInternational Journal of Biological Macromolecules 182 (2021) 264and tumor suppressor genes (TSGs) can trigger the development of tumors. The majority of approved targeted drugs are designed to inhibit the oncogenic activities of oncogenes. Gefitinib and erlotinib are small molecule inhibitors of epidermal growth factor receptor tyrosine kinase (EGFR-TK) [1], imatinib is a BCR-ABL tyrosine kinase inhibitor [2], and some monoclonal antibodies act against certain oncogenes [3]. On the other hand, DNA methylation inhibitor azacitidine (AZA) and histone deacetylase inhibitors trichostatin A (TSA) can upregulate the global expression levels of TSGs. TSG DLC1 plays crucial roles in the genesis and development of a variety of tumors. DLC1 is a Rho GTPase-active protein (RhoGAP) that negatively regulates Rho proteins, which play critical roles in several cellular processes, including proliferation, polarity, motility, cytoskeletal remodeling, morphogenesis and vesicular trafficking [4]. Loss of heterozygosity of DLC1 was first detected in the primary tumors of hepatocellular carcinoma (HCC) and HCC cell lines in 1998 [5]. Lahoz and Hall asserted that "DLC1 is a human tumor suppressor that is not only deleted in liver cancer but also deleted in lots of cancers" [6]. DLC1 belongs to the DLC family, which contains two other members, DLC2 and DLC3. DLCs are structurally and functionally similar to each other [70]. They all possess three functional domains, sterile  motif (SAM) domain, catalytic RhoGAP domain and steroidogenic acute regulatory protein (StAR)-related lipid transfer (START) domain, and a serine-rich region (SR region) or a linker region located between the SAM and RhoGAP domains [11] (Fig. 1). Functionally, consistent with the conservation of the RhoGAP domain, DLCs show strong GAP activity toward RhoA and weak activity for Cdc42 but not for Rac [10,12,13]. Overexpression of all DLCs causes a similar GAPdependent phenotype, characterized by a rounded cell morphology with long protrusions and the loss of actin stress fibers [13,14]. Although  Carrier Free Antibodies  are downregulated in most cancer cells and tumor tissues, DLC1 is thought to be the principal biologically relevant downregulated DLC family member [15], and the mechanisms of the other two members on tumor inhibition are not well explained. DLC1 has five isoforms, DLC1 isoforms 1 [168]. Among them, DLC1 isoform 1, which has the largest protein product, is often used for coding sequences in most large databases [19]. Most studies have focused on the widely distributed and predominant DLC1 isoform 2 [17]. In this review, we summarize the anticancer effects of DLC1 isoform 2 and regulation of DLC1 expression, localization and modification. We also summarize the drugs or compounds that targeted DLC1 for anticancer target therapy. 2. DLC1 is a tumor suppressor DLC1 plays an indispensable role in restraining tumor cell development in solid tumors [15,20,21]. First, DLC1 inhibits the migration and invasion of tumor cells [229]. During cell migration, enhanced Rho signaling facilitates the formation of lamellipodia at the leading edge.DLC1 inhibits migration by downregulating Rho signaling [30]. Interestingly, simultaneous downregulation of DLC1 and Rho effector protein Dia1, the mammalian ortholog of Drosophila diaphanous 1, completely abrogated cell migration resulting from DLC1 knockdown, indicating that DLC1 controls cell migration by modul.
